5-Amino-1MQ (5-amino-1-methylquinolinium) is a cell-permeable, selective small molecule inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). NNMT is a cytosolic enzyme that plays a critical role in cellular energy homeostasis by catalyzing the methylation of nicotinamide, which can lead to the depletion of intracellular NAD+ levels. By inhibiting NNMT, 5-Amino-1MQ facilitates the preservation of NAD+ pools and increases the activity of the sirtuin-1 (SIRT1) pathway. Research applications include investigations into metabolic regulation, white adipose tissue signaling, muscle stem cell (satellite cell) regeneration, and the modulation of energy expenditure in models of obesity and metabolic dysfunction.
